چکیده

در این تحقیق اثر آنتی اکسیدانی عصاره برگ زیتون در مقایسه با ویتامین­های C و E در اسپرم سرد شده گاو در طی مدت   48ساعت نگهداری در شرایط دمایی پنج درجه
سانتیگراد مقایسه شد. اسپرم از قسمت دم اپیدیدیم بیضه 10رأس گاو نر نژاد هولشتاین دورگ با میانگین سنی سه تا پنج سال جمع­آوری و با افزودن رقیق­کننده تخم­مرغ و
سیترات سدیم حاوی عصاره برگ زیتون (در سه غلظت نیم، یک و دو میلی­گرم در میلی­لیتر) و ویتامین­های E  و C (هرکدام با غلظت دو میلی­گرم در میلی­لیتر) رقیق شدند. افزودن
عصاره برگ زیتون به رقیق­کننده، تحرک اسپرم را کاهش داد (05/0P<). سلامت غشا اسپرم گاو استفاده از ویتامین E بالاتر از سایر تیمارها بود (05/0P<). میزان پراکسیداسیون چربی­ها در تیمار حاوی دو میلی گرم در میلی لیتر عصاره برگ زیتون کمتر از سایر تیمارها بود (05/0P<). نتایج این تحقیق نشان داد که افزودن عصاره برگ زیتون به رقیق­کننده اسپرم در طی مدت 48 ساعت نگهداری در شرایط دمایی پنج درجه سانتیگراد علیرغم کاهش میزان اکسیداسیون لیپیدی موجب کاهش تحرک اسپرم گاو می­شود و
بنابراین ترکیب مناسبی برای استفاده در رقیق­کننده­های اسپرم گاو نمی­باشد.

In this study antioxidant effects of olive leaf extract (OLE) in comparison to vitamin C and E on chilled-stored (at 5 °C) bovine sperm was evaluated. The sperm samples collected from epididymis of ten Holstein bovine testis with 3-5 years old, and were diluted with egg yolk and sodium citrate containing 0.5, 1.0 and 2.0 mg/ml of OLE as well as 2 mg/ml vitamin E and C. Addition of OLE significantly reduced sperm motility (P<0.05). Vitamin E significantly improved sperm membrane integrity compared to the other groups (P<0.05). TBARS in 2 mg/ml of OLE treatment was significantly less than that in other treatments (P<0.05). The results showed that addition of OLE to bovine sperms stored at 5 ͦ C for 48 h negatively affected sperm motility despite of a decrease in lipid peroxidation, and therefore, it is not suitable extender to be used for bovine spermatozoa.
