چکیده

فاکتور‌های نسخه برداری با اتصال به نواحی پروموتوری ژن‌ها و تنظیم بیان آن‌ها نقش مهمی در کنترل باروری حیوانات دارند. در این مطالعه، بیان ژن‌های nobox، ovol1 و zp3 که از ژن‌های کاندید درگیر در مسیر‌های تنظیمی ژن‌ها می‌باشند، در دو گروه چند قلوزا و تک قلوزای گوسفندان شال بررسی شد. تعداد شش رأس گوسفند با توجه به شجره آنها از گله اصلاح شده ایستگاه شال قزوین جدا شده و برای این تحقیق مورد استفاده قرار گرفتند. پس از همزمان سازی فحلی، گوسفندان با روش بیهوش عمومی جراحی شده و فولیکول‌های بالغ تخمدان با روش آسپیره کردن استحصال شدند. پس از استخراج RNA، cDNA نمونه‌ها ساخته شد. برای بررسی بیان ژن‌های فوق، آغازگر‌های اختصاصی هر ژن طراحی شده و سپس با استفاده از روشReal-Time PCR و با بکارگیری روش ΔΔCt بیان نسبی ژن‌ها سنجیده شد. مقایسات بین میانگین ها، با استفاده از رویه آماری T-Test انجام شد. همچنین با استفاده از نرم‌افزار BDGP نواحی پروموتوری و فاکتور‌های نسخه‌برداری ژن‌های مورد مطالعه بررسی شد. بیان ژن‌های nobox و ovol1 در گروه حیوانات چندقلوزا، معنی‌دار بود (P

چکیده [English]

Transcription factors have important role in controls of animal fertility by binding to promoter regions of genes and regulation of their expression. In this study, nobox, ovol1 and zp3 genes expression, that are candidate genes involved in regulatory pathways of genes, in two categories of uniparous and multiparous Shall sheep were investigated. According to the pedigree of animals, a totally of six ewes selected from the herd of Qazvin Shall breeding center and applied for this experiment. After estrus synchronization, ewes were operated by general anesthesia technique and ovarian mature follicles were extracted by aspiration. After RNA extraction, sample’s cDNA was constructed. To investigation the expression of the above genes, a set of specific primers was designed for a gene, and then relative expression of these genes were measured using Real-Time PCR method and by employment of ∆∆Ct approach.
