چکیده

هدف: ارتقای کیفیت تغذیه‌ای تخم‌مرغ از طریق غنی‌سازی با ریزمغذی‌هایی مانند سلنیوم، یک راهبرد ارزش‌آفرین در صنعت طیور است. تخم‌مرغ غنی‌شده با سلنیوم نه تنها می‌تواند به تأمین این عنصر ضروری در رژیم غذایی انسان کمک کند، بلکه ممکن است از طریق بهبود پروفایل آنتی‌اکسیدانی خود، به پایداری اکسیداتیو و ماندگاری بهتر محصول نیز بینجامد. علاوه بر این، نقش سلنیوم در تنظیم بیان ژن‌های درگیر در مسیرهای پاسخ به استرس اکسیداتیو و سم‌زدایی، لایه دیگری از تأثیرات مثبت این عنصر را بر سلامت سلولی آشکار می‌سازد. با توجه به تفاوت‌های ذاتی منابع مختلف سلنیوم در جذب، متابولیسم و کارایی زیستی، ارزیابی دقیق تأثیر این منابع بر شاخص‌های عملکردی، سلامت و کیفیت محصول در مرغ تخم‌گذار ضروری است. بنابراین، هدف از این مطالعه، مقایسه اثرات مکمل‌سازی جیره با دو منبع رایج سلنیوم (سلنیت-سدیم و سلنیوم-متیونین) در دو سطح متفاوت بر عملکرد تولیدی، شاخص‌های استرس اکسیداتیو، تجمع سلنیوم در تخم‌مرغ و کبد، و همچنین بیان ژن‌های کلیدی آنتی‌اکسیدانی در کبد مرغ‌های تخم‌گذار بود.

روش پژوهش: آزمایشی با استفاده از 300 قطعه مرغ تخم‌گذار لگهورن سویه های- لاین ، در قالب طرح کاملا تصادفی با استفاده از 5 تیمار، 4 تکرار و 10 قطعه جوجه در هر تکرار اجرا شد. تیمارهای آزمایشی شامل: 1- جیره پایه (فاقد سلنیوم)، 2- جیره پایه+ mg/kg5/0 سلنیت- سدیم، 3- جیره پایه + mg/kg 1 سلنیت- سدیم، 4- جیره پایه + mg/kg 5/0 سلنیوم- متیونین، 5- جیره پایه + mg/kg 1 سلنیوم- متیونین بودند. صفات عملکردی شامل مصرف خوراک، افزایش وزن بدن و ضریب تبدیل خوراک، به صورت دوره‌ای اندازه‌گیری شد و فعالیت آنتی اکسیدانی، بیان ژن GPx و TXNDR1 و میزان ذخیره-شوندگی سلنیوم در زرده و سفیده تخم‌مرغ و کبد مورد ارزیابی قرار گرفت. تجزیه و تحلیل آماری داده‌ها توسط نرم افزار SAS انجام شد.

یافته‌ها: نتایج نشان داد مصرف منابع مختلف سلنیوم آلی و غیرآلی در هیچ یک از دوره‌‌های تولید بر میانگین افزایش وزن، خوراک مصرفی و ضریب تبدیل خوراکی در مرغ‌های تخم‌گذار تاثیر معنی‌داری نداشت (05/0<P). همچنین فعالیت آنتی‌اکسیدانی و میزان مالون‌دی‌آلدهید خون در مرغ‌های تخم‌گذار تحت تأثیر سطوح مختلف منابع سلنیوم قرار نگرفت (05/0<P). در عین حال نتایج نشان داد مصرف سلنیوم منجر به کاهش معنی‌دار میزان مالون‌دی‌آلدهید کبد در مقایسه با تیمار شاهد شد (05/0P<)؛ تیمارها ی دریافت کننده سلنیوم آلی در مقایسه با سلنیوم غیرآلی به شکل معنی‌داری میزان تولید مالون‌دی‌آلدهید کبدی پایین‌تری از خود نشان دادند (05/0P<). مصرف یک میلی‌گرم در کیلوگرم سلنیوم آلی به شکل معنی‌داری منجر به ذخیره بیشتر سلنیوم در زرده و سفیده تخم‌مرغ خشک‌شده در مقایسه با سایر تیمارهای آزمایشی شد (05/0P<). مصرف سلنیوم به شکل معنی‌داری منجر به افزایش غلظت سلنیوم در زرده و سفیده تخم‌مرغ تازه می‌شود (05/0P<) و به‌شکل معنی‌داری منجر به افزایش غلظت سلنیوم در کبد شد (05/0P<). تیمارهای دریافت‌کننده سلنیوم آلی در مقایسه با تیمارهای دریافت کننده سلنیوم معدنی میزان بیان ژن‌های GSH و TXNDR1 بالاتری از خود نشان دادند (05/0P<).

نتیجه‌گیری: بر اساس یافته‌های به‌دست‌آمده اگرچه سلنیوم بر شاخص های عملکردی تاثیر معنی‌داری نداشته اما مصرف یک میلی‌گرم در کیلوگرم سلنیوم-متیونین در جیره باعث کاهش میزان مالون‌دی‌آلدهید، افزایش میزان سلنیوم در تخم‌مرغ و تنظیم مثبت بیان ژن‌های آنتی‌اکسیدانی شود و می تواند به عنوان یک راهکار تغذیه‌ای موثر برای ارتقای سلامت پرنده و بهبود کیفیت تغذیه‌ای پیشنهاد شود.

objective: Enhancing the nutritional quality of eggs through enrichment with micronutrients such as selenium is a value-adding strategy in the poultry industry. Selenium-enriched eggs can not only help supply this essential element in the human diet but may also lead to improved oxidative stability and longer shelf life of the product due to their enhanced antioxidant profile. Furthermore, selenium's role in regulating the expression of genes involved in oxidative stress response and detoxification pathways reveals another layer of its positive effects on cellular health. Given the inherent differences among various selenium sources in absorption, metabolism, and bioavailability, a precise evaluation of their impact on performance, health, and product quality in laying hens is essential. Therefore, this study aimed to compare the effects of dietary supplementation with two common selenium sources (sodium selenite and selenomethionine) at two different levels on production performance, oxidative stress indicators, selenium accumulation in eggs and liver, and the expression of key antioxidant genes in the liver of laying hens.

Method: An experiment was conducted using 300 Hy-Line W-36 laying hens in a completely randomized design with 5 treatments, 4 replicates, and 10 birds per replicate. The experimental treatments included: 1) Basal diet (without selenium), 2) Basal diet + 0.5 mg/kg sodium selenite, 3) Basal diet + 1 mg/kg sodium selenite, 4) Basal diet + 0.5 mg/kg selenomethionine, and 5) Basal diet + 1 mg/kg selenomethionine. Performance traits, including feed intake, body weight gain, and feed conversion ratio, were measured periodically. Antioxidant activity, expression of GPx and TXNRD1 genes, and selenium deposition in egg yolk, egg albumen, and liver were evaluated. Statistical analysis of the data was performed using SAS software.

Results: The results showed that supplementation with different organic and inorganic selenium sources had no significant effect on average weight gain, feed intake, or feed conversion ratio in laying hens during any production period (P>0.05). Similarly, plasma antioxidant activity and malondialdehyde (MDA) levels were not significantly affected by different selenium sources (P>0.05). However, selenium supplementation led to a significant decrease in hepatic MDA levels compared to the control group (P<0.05). Treatments receiving organic selenium showed significantly lower hepatic MDA production compared to those receiving inorganic selenium (P<0.05). Supplementation with 1 mg/kg organic selenium resulted in significantly higher selenium deposition in dried egg yolk and albumen compared to other experimental treatments (P<0.05). Selenium supplementation significantly increased selenium concentration in fresh egg yolk and albumen (P<0.05) and significantly elevated selenium concentration in the liver (P<0.05). Treatments receiving organic selenium showed higher expression levels of GSH and TXNRD1 genes compared to those receiving inorganic selenium (P<0.05).

Conclusions: Based on the findings, although selenium did not significantly affect performance indicators, dietary supplementation with 1 mg/kg selenomethionine reduced MDA levels, increased selenium content in eggs, and positively regulated the expression of antioxidant genes. Therefore, it can be recommended as an effective nutritional strategy to enhance bird health and improve the nutritional quality of eggs.
