تعیین سطح مطلوب انرژی و پروتئین در جیره رشد بلدرچین ژاپنی (Coturnix coturnix japonica)
بهمنظور تعیین سطح مطلوب انرژی و پروتئین در جیرۀ دورۀ رشد بلدرچین ژاپنی، از تعداد 360 قطعه بلدرچین ژاپنی یکروزه در آزمایش فاکتوریل 3 × 3، [سه سطح انرژی سوختوسازپذیر (2750، 2850 و 2950 کیلوکالری در کیلوگرم) و سه سطح پروتئین (24، 26 و 28 درصد)] در قالب طرح کاملاً تصادفی با چهار تکرار و 10 پرنده در هر تکرار استفاده شد. مصرف خوراک و افزایش وزن هفتگی اندازهگیری، و ضریب تبدیل غذا محاسبه شد. در پایان دوره، از هر واحد آزمایشی چهار قطعه پرنده (از هر دو جنس) انتخاب و پس از خونگیری برای بررسی صفات لاشه، کشتار شدند. غلظت سرمی هورمون رشد در نمونههای خون اندازهگیری شد. افزایش وزن روزانه، ضریب تبدیل، و بازده لاشۀ پرندگان تیماری که جیرۀ حاوی 2850 کیلوکالری در کیلوگرم انرژی سوختوسازپذیر و 26 درصد پروتئین دریافت کردند بهتر بود (05/0>P). غلظت سرمی هورمون رشد پرندگان تغذیهشده با جیرۀ حاوی 2750 کیلوکالری در کیلوگرم انرژی سوختوسازپذیر بیشتر بود (05/0>P). بدین ترتیب جیرۀ حاوی 2850 کیلوکالری در کیلوگرم انرژی سوختوسازپذیر و 26 درصد پروتئین بهعنوان جیرۀ رشد بلدرچین توصیه میشود.

اثر سطوح متفاوت مس و کاسنی بر کلسترول زرده و عملکرد مرغهای تخمگذار
این پژوهش بهمنظور بررسی تأثیرات سطوح متفاوت گیاه کاسنی و مس بر عملکرد و کلسترول زرده در مرغهای تخمگذار انجام شد. تعداد 324 قطعه مرغ تخمگذار سویههای لاین w-36از 38 تا 50 هفتگی در قالب طرح بلوکهای کاملاً تصادفی با نه تیمار، سه تکرار، و 12 قطعه در هر تکرار بهمدت 12 هفته بررسی شد. تیمارهای آزمایشی شامل شاهد، چهار سطح سولفات مس (150، 200، 250، و 300 میلیگرم در کیلوگرم جیرۀ غذایی) و چهار سطح کاسنی (15، 20، 25، و 30 درصد جیرۀ غذایی) بود. نتایج نشان داد که میانگین کلسترول سرم و زردۀ تخممرغ در تیمارهای مس و کاسنی از تیمار شاهد کمتر بود (05/0P<). در تیمار 300 میلیگرم مس، کلسترول سرم و زردۀ تخممرغ کمتر بود (05/0P<)، ولی وزن تخممرغ و درصد تولید در مقایسه با شاهد کمتر و ضریب تبدیل غذا بیشتر بود (05/0P<). درصد تولید در تیمار 25 و 30 درصد کاسنی در مقایسه با تیمار شاهد کمتر بود (05/0P<). در تیمارهای سطوح گوناگون کاسنی، رنگ زرده در مقایسه با تیمار شاهد و مس بیشتر بود (05/0P<). باتوجه به نتایج تحقیق حاضر، مقادیر 150 میلیگرم مس در کیلوگرم جیره یا 15 درصد کاسنی در جیره برای کاهش کلسترول زردۀ تخممرغ توجیهپذیر است.

بررسی تعداد سلولهای سوماتیک شیر در گاوداریهای صنعتی و سنتی استان تهران
در این تحقیق، تعداد سلولهای سوماتیک شیر در شش گاوداری صنعتی و سنتی استان تهران در فصول گوناگون سال بررسی شد. برمبنای تعداد سلولهای سوماتیا، میزان فراوانی ورم پستان براورد شد. تعداد 32610 رکورد بااستفاده از رویههای متفاوت نرمافزار آماری SAS تجزیه و تحلیل شد. میانگین حداقل مربعات سلول سوماتیک (تعداد ×1000 در هر میلیلیتر) ± خطای معیار در روش پرورشی صنعتی و سنتی بهترتیب 1/80(60/12±) و 6/234(97/12±) براورد شد. تعداد سلولهای سوماتیک شیر در گاوداریهای سنتی حدود 9/2 برابر گاوداریهای صنعتی بود (05/0P<). بیشترین تعداد سلول سوماتیک شیر در فصل تابستان بود و تفاوت آن با دو فصل بهار و پاییز معنیدار بود (05/0P<). برمبنای تعداد سلولهای سوماتیک، درصد ورم پستان تحت بالینی و بالینی در روش پرورش صنعتی بهترتیب 6/31 و 8/11 درصد براورد شد. مقادیر مشخصشده برای روش پرورش سنتی 6/59 و 7/34 درصد بود. نتایج این مطالعه نشان داد اگر تعداد سلولهای سوماتیک شیر به نصف کاهش یابد، از میزان وقوع ورم پستان 30-50 درصد میکاهد.

اثر افزودن یونجه خشک به عنوان جاذب ر طوبت بر کیفیت تخمیر و تولید پساب در سیلاژ ذرت
هدف از انجام مطالعة حاضر، بررسی اثر یونجة خشک (به عنوان جاذب رطوبت) بر تولید پساب و کیفیت تخمیر سیلاژ ذرت بود. در طرحی کاملاً تصادفی در قالب فاکتوریل 4×3 به سه علوفة کامل ذرت با مادة خشک 3/18، 7/21 و 5/25 درصد، چهار سطح جاذب صفر، 5، 10 و 20 درصد اضافه و در سه تکرار در لولههایی از جنس پلیوینیل کلریدی با عنوان سیلوی کوچک به مدت 90 روز سیلو شدند. با افزایش سطح استفاده از جاذب، مقادیر خاکستر و نیتروژن کل سیلاژها افزایش و الیاف نامحلول در شویندة خنثی به طور معنیداری کاهش یافت (05/0>P). اثر سطوح جاذب و رطوبت روی مادة خشک سیلاژها معنیدار بود و مادة خشک با 10 و 20 درصد جاذب بیشترین افزایش را نشان داد (05/0>P). تمام سیلاژها دارای pH مطلوب (در محدودة 79/3 تا 33/4) بودند. با وجود این، pH سیلاژها تحت تأثیر اثر متقابل رطوبت و سطح جاذب قرارگرفت (05/0>P). با افزایش سطح استفاده از جاذب، pH سیلاژها افزایش یافت اما این افزایش در سیلاژهایی با 26 درصد مادة خشک با شدت کمتری مشاهده شد. همچنین، مقادیر کربوهیدرات محلول باقیمانده تحت تأثیر اثر متقابل بین رطوبت و سطح جاذب قرارگرفت. بدین صورت که در سیلاژهای با 18 درصد مادة خشک اختلاف بین سطوح جاذب کمتر از اختلاف آنها در سیلاژهای با 22 و 26 درصد مادة خشک بود (05/0P<). با افزایش سطح یونجة خشک، پایداری هوازی در سیلاژهایی که رطوبت اولیة بالاتری داشتند بهبود یافت و به موازات افزایش مادة خشک اولیة علوفة ذرت، اختلاف در پایداری هوازی کمتر شد (05/0>P). با افزایش سطح یونجه در سیلاژ، تولید پساب کاهش یافت، بهطوریکه پساب در سیلاژهای حاوی 20 درصد یونجه خشک به صفر رسید (05/0>P). در پساب سیلاژهایی که سطوح بالاتری جاذب داشتند غلظت مادة خشک و نیتروژن بیشتر بود، اما کل مادة خشک و نیتروژن هدررفته در این تیمارها کاهش یافت. نتایج نشان داد استفاده از حداقل 10 درصد یونجة خشک باعث کاهش مؤثر پساب، بهبود پایداری هوازی و تولید ذرت سیلوشده با pH مطلوب میگردد.

پاسخ ایمنی جوجه های گوشتی تحت تنش سرمایی به ویتامین C و کوآنزیم Q10
این آزمایش بهمنظور بررسی نقش آنتیاکسیدانی ویتامین C (VC) و کوآنزیم Q10 (CoQ10) بر پاسخ ایمنی جوجههای گوشتی تحت شرایط تنش سرمایی بااستفاده از تعداد 500 قطعه جوجۀ نر آرین در قالب طرح کاملاً تصادفی با پنج تیمار و پنج تکرار انجام شد. تیمارهای آزمایشی شامل گروه کنترل مثبت (PC، شرایط عادی پرورش، و بدون دریافت آنتیاکسیدان)، گروه کنترل منفی (NC، تنش سرمایی، و بدون دریافت آنتیاکسیدان)، تنش سرمایی بههمراه تیمارهای حاوی VC (300 میلیگرم در کیلوگرم جیره)، CoQ10 (40 میلیگرم در کیلوگرم جیره)، و مخلوط VC+CoQ10 با دزهای ذکرشده در جیرۀ پایه بود. برای اعمال تنش سرمایی، از 15 تا 42 روزگی، دمای سالن 15 درجۀ سانتیگراد ثابت نگهداشتهشد. صفات تیتر واکسنها، پاسخ ایمنی هومورال، ایمنی سلولی، و وزن نسبی اندامهای ایمنی مطالعه شد. نتایج نشان داد که با وجود کاهش وزن نسبی طحال تحت تنش سرمایی، تفاوتها معنیدار نبودند. پرندگانی که تحت تأثیر تنش سرمایی قرار گرفتند، وزن نسبی بورس بالاتری داشتند (01/0P<) و استفاده از آنتیاکسیدانها بهویژه CoQ10 وزن نسبی آنها را کاهش داد (05/0>P). درصد لنفوسیتها درنتیجۀ تنش سرمایی، کاهش یافت (05/0>P). بهبود پاسخ ایمنی سلولی هم در پاسخ به تزریق زیرجلدی فیتوهماگلوتینین و هم در تکثیر لنفوسیتهایT در شرایط آزمایشگاهی مشاهده گردید (05/0P<). نتایج این تحقیق نشان داد که استفاده از مواد آنتیاکسیدانی VC و CoQ10 در شرایط تنش سرمایی باعث بهبود سیستم ایمنی و کاهش تلفات شد.

تعیین پاسخ بوقلمونهای در حال رشد به سطوح لیزین هضمشدنی ایلئومی استاندارد شده با استفاده از روشهای گوناگون آماری
بهمنظور تعیین مناسبترین سطح لیزین هضمشدنی ایلئومی استانداردشده در مرحلۀ رشد (49-28 روزگی) بوقلمونهای نر گوشتی، 160 قطعه بوقلمون در قالب طرح کاملاً تصادفی با هشت تیمار (هشت سطح لیزین هضمشدنی ایلئومی استانداردشده، 15/1 (جیرۀ پایه)، 225/1، 3/1، 375/1، 45/1، 525/1، 6/1، و 675/1 درصد) در چهار تکرار و پنج قطعه پرنده در هر تکرار استفاده شدند. افزایش سطوح لیزین هضمشدنی باعث افزایش وزن بدن و کاهش ضریب تبدیل غذایی گردید. گروه آزمایشی حاوی 6/1 درصد لیزین هضمشدنی، بیشترین افزایش وزن بدن و کمترین ضریب تبدیل غذایی را داشت که با جیرۀ پایۀ (15/1 درصد لیزین هضمشدنی) تفاوت معنیداری نشان داد (05/0P<). با برازش مدلهای رگرسیون خط شکسته، منحنی درجۀ دو، و خط شکستۀ درجۀ دو میزان نیاز لیزین هضمشدنی، بهترتیب برای افزایش وزن بدن 11/0 ± 47/1، 04/0 ± 57/1 و 05/0 ± 49/1 درصد تخمین زده شد. نیاز لیزین هضمشدنی بااستفاده از مدلهای فوق برای بهترین ضریب تبدیل غذایی بهترتیب 17/0 ± 57/1، 02/0 ± 61/1، و 08/0 ± 59/1 درصد تعیین شد. باتوجه به نتایج تحقیق حاضر، نیاز لیزین هضمشدنی بوقلمونهای نر گوشتی در دورۀ رشد برای افزایش وزن بدن در دامنۀ 47/1 تا 57/1 و برای ضریب تبدیل غذایی در دامنۀ 57/1 تا 61/1 درصد توصیه میشود.

مقایسه روشهای گوناگون ارزیابی ژنومیک در صفاتی با معماری ژنتیکی متفاوت
هدف از انجام تحقیق حاضر، مقایسۀ روشهای گوناگون آماری در پیشبینی ارزشهای اصلاحی ژنومیک برای صفاتی با معماری ژنتیکی متفاوت ازنظر توزیع تأثیرات ژنی و نیز تعداد متفاوت جایگاههای صفت کمّی (QTLs) بااستفاده از شبیهسازی کامپیوتری است. بدین منظور، ژنومی حاوی 500 نشانگر تکنوکلئوتیدی دوآللی (SNP) روی کروموزومی به طول 100 سانتیمورگان شبیهسازی، و توزیعهای متفاوت تأثیرات ژنی (یکنواخت، نرمال، و گاما) و نیز سه تعداد QTL (50، 100 و 200) بهصورت فرضیههای شبیهسازی صفت روی آن درنظر گرفته شد. بدین ترتیب نه صفت با معماری ژنتیکی متفاوت ایجاد شد. بهمنظور پیشبینی ارزشهای اصلاحی افراد موجود در جمعیتهای مرجع و تأیید، از شش روش بهترین پیشبینی نااریب خطی ژنومیک[1] (GBLUP)، رگرسیون ریدج[2] (RRBLUP)، بیز A (BayesA)، بیز B (BayesB)، بیز Cπ (BayesCπ)، و بیز L (BayesL) استفاده گردید. با افزایش فاصلۀ بین نسل مرجع و نسل تأیید ناشی از برهمخوردن فاز لینکاژ، صحت ارزشهای اصلاحی ژنومیک در تمام روشها کاهش معنیداری نشان داد (05/0P<). همچنین زمانیکه توزیع تأثیر ژنی گاما بود روشهای بیزی در مقایسه با روش GBLUP و RRBLUP برتری آشکاری نشان دادند. هنگامیکه توزیع تأثیرات ژنی نرمال بود، بیشترین صحت ارزشهای اصلاحی بهدست آمد. با افزایش تعداد QTLها بهسمت 200، صحت ارزشهای اصلاحی ژنومیک کاهش یافت. نتایج این تحقیق نشان داد که درمجموع روشهای بیزی و GBLUP ازنظر صحت ارزشهای اصلاحی پیشبینیشده در مقایسه با روش RRBLUP عملکرد بهتری دارند. نتایج تحقیق حاضر نشان میدهد هنگامیکه معماری صفات بررسیشده از مدل تعداد زیاد جایگاه ژنی پیروی نکند، معمولاً روشهای بیزی بر روشهای GBLUP و RRBLUP ارجحیت دارند. 1.
